I believe you are asking if you can log into persistence, run your updates or install nessus, then be able to access those update programs/nessus in a non-persistence mode so you can reverse anything that may mess up by rebooting.
That would be excellent. However, persistence runs from the separate partition you made on your stick whereas non-persistence does not. So when you run it in non-persistence, it doesn't tap into the persistent partition you created thus you wont be able to use nessus or updated programs.
